{"id":368,"date":"2014-03-27T17:28:01","date_gmt":"2014-03-27T16:28:01","guid":{"rendered":"http:\/\/s519078666.onlinehome.fr\/?page_id=368"},"modified":"2015-03-10T15:28:36","modified_gmt":"2015-03-10T14:28:36","slug":"la-therapie-genique-gene-abca4","status":"publish","type":"page","link":"http:\/\/www.stargardt.fr\/test\/recherche-et-etudes-en-cours\/la-therapie-genique-gene-abca4\/","title":{"rendered":"Th\u00e9rapie g\u00e9nique"},"content":{"rendered":"<h2>Restaurer les fonctions du g\u00e8ne ABCA4<\/h2>\n<p><strong>Dans le traitement des maladies orphelines, le but de la th\u00e9rapie g\u00e9nique est d\u2019introduire chez une personne porteuse de la mutation g\u00e9n\u00e9tique, la copie normale du g\u00e8ne ou des g\u00e8nes alt\u00e9r\u00e9 responsable de ce trouble afin de remplacer le(s) g\u00e8ne(s) d\u00e9ficient(s).<\/strong><\/p>\n<p>Dans la maladie de Stargardt, l\u2019objectif recherch\u00e9 de la th\u00e9rapie g\u00e9nique est d\u2019introduire l\u2019ADN sain (ou normal) du g\u00e8ne ABCA4 dans les cellules de la r\u00e9tine afin de restaurer les fonctions normales de la prot\u00e9ine ABCA4 et pr\u00e9venir la d\u00e9gradation de la r\u00e9tine. Pour\u00a0introduire ce g\u00e8ne sain dans les cellules de la r\u00e9tine, les chercheurs utilisent des vecteurs qui vont p\u00e9n\u00e9trer dans les cellules et infiltrer l\u2019ADN sain dans leur noyau. Le g\u00e8ne sain se trouvant alors dans le noyau,\u00a0 peut produire la prot\u00e9ine normale. Les vecteurs utilis\u00e9s peuvent \u00eatre des virus dont toute activit\u00e9 infectieuse virale a \u00e9t\u00e9 retir\u00e9e (r\u00e9trovirus, ad\u00e9novirus, lentivirus).<\/p>\n<p>Une des contraintes du g\u00e8ne ABCA4 est li\u00e9e \u00e0 sa taille\u00a0: c\u2019est un g\u00e8ne relativement long et lourd.
